Abstract

The primary objective of the proposed work is to determine the bluetongue virus (BTV) structure of VP7 and VP3 interactions by analysis of BTV core-like particles (CLPs) as surrogate of viral core particles. The CLPs can be synthesised in large quantities by expressing VP7 and VP3 using baculovirus expression vectors will be crystallised and analysed by X-ray diffraction on the available 3-dimensional structure of BTV VP7. The structure of VP7 and CLPs under alternative conditions (various pH etc.) and by cryo-electron microscopy as well as FAB-CLP complexes will also be analysed. In addition, the structural analysis of two minor proteins (VP6 and VP4) of the core will also be initiated.
