Abstract

Funding is requested to establish a stopped flow facility capable of recording in absorbance, fluorescence and circular dichroism (c.d.) modes. This will be used to provide kinetic insights into the key biological processes of protein folding and assembly and macromolecule/ligand interaction. Four systems (which are ideal for this work) have been chosen to illustrate these themes:- (1) The folding of the monomeric phosphoglycerate mutase from Schizosaccharomyces pombe, (2) The folding and assembly of the type II dehydroquinase from Streptomyces coelicolor, (3) The folding and reactivation of BM3 cytochrome P450 from Bacillus megaterium, and (4) The interaction between DNA methyltransferases and their cognate DNAs.
