Abstract
We aim to describe the folding pathway of a small beta-sandwich protein by a combination of kinetic and structural methods. We will define: (1) the rates of formation and decay of intermediates, (2) the conformational properties and stabilities of key states in the process and (3) the residue contacts responsible for guiding the pathway. X-ray crystallography, mutational analysis, protein sub-fragments, proton-exchange NMR and transient kinetics will be used to elucidate the mechanism.
