Abstract

This project aims to determine the structures of a transmembrane in lipid bilayers using a non-crystallographic and non-perturbing method, namely novel solid state NMR. The peptide chosen can adopt an alpha-oligomeric or beta-oligomeric form, as resolved by CD, FTIR and Raman spectroscopy, depending upon conditions used for incorporation into bilayers. Intra- atomic distances between selected pairs of labelled (13C) carbon atoms in a bilayer-embedded peptide will be measured to resolve secondary structure dimensions and test indirect structural prediction methods. Intermolecular distances will permit helix-helix distances and associations to be assessed.
