Abstract
1. Employ photochemical crosslinking, chemical protection and mutagenesis to identify amino acids in the NF-kB transcription factor that are in direct contact with the DNA binding site and evaluate their contribution to recognition. 2. Use partial proteolysis, chemical protection and mutagenesis to define how individual residues, present at the interface between NF-kB and the inhibitor protein IkB, contribute to molecular recognition. 3. Using appropriately labelled NF-kB subunits use fluorescence energy transfer measurements to establish how IkB inhibits NF-kB DNA binding activity.
