Abstract
Two recent developments will allow us to investigate intra- and inter-protein electron transfer to a new level of detail, using flavocytochrome b2 as an experimental system. Firstly, the modelling of a realistic electron transfer complex between four molecules of cytochrome c and a flavocytochrome b2 tetramer presents us with a testable model for protein-protein interactions that promote complex formation. This will form the basis of the major part of the work proposed here. Secondly, the recent determination of crystal structures of mutant forms of flavocytochrome b2 demonstrates the ability to analyse structure-function correlations in fine molecular detail and we shall analyse intra-protein electron transfer in flavocytochrome b2 in relation to the structures of these two mutant structures which have alterations in the inter-domain interface.
