Abstract

Our overall aim is to understand the pathway and mechanism of electron transfer in photosystem 2 (PS2), a photosynthetic membrane-protein complex found in plants, algae and cyanobacteria. It is responsible through water oxidation for most of our oxygen atmosphere and is an important part of the biological solar energy conversion system. In previous work, key amino acid sites in the core complex were predicted by molecular modelling and specific mutants constructed in several laboratories. We will now investigate these by spectroscopic techniques and compare them to the native state which we have characterised. This project will give fine structural information on cofactor positions, H-bonding, electron spin density of cofactors and how their redox properties are controlled. Together these will provide a model of electron transfer through PS2, a crucial aid to future experiments to mimic or modify photosynthetic systems.
