Abstract
We aim to improve our understanding of the principles underlying the folding of beta sandwich proteins. This will involve a range of experimental approaches using a small protein exemplifying this structural class, tendamistat, as a model system. Our strategy will include (i) study of the feasibility of alternative strand topologies and their significance for folding, through protein redesign experiments; (ii) detailed investigation by NMR and protein engineering of the conformational properties of large fragments of the protein, some of which appear to have embryonic structure.
