Abstract

The proposal is for research into methods for improving electron density maps of macromolecular crystals based on density modification, symmetry averaging and other constraints on the phases. This project is aimed specifically at improving the SQUASH program, which has been developed in York and which applied a combination of phase constraints simultaneously. New features include the use of critical points in the electron density map to obtain molecular skeletons; use of the skeleton in density modification and in rapid translation and rotation searches and the use of improved figures of merit in an iterative procedure to increase the entropy of the map.
