Abstract

We aim to determine the three-dimensional structure of a Class II fructose-1,6-bisphosphate aldolase using crystallographic methods and to characterise fully the structure-activity relationships of this enzyme. To then utilise protein engineering methodology to redesign the enzyme's active site so creating new aldolases with designed specificity. Within the remit of this proposal we also plan to clone and overexpress the Class I aldolase of E. coli for comparative studies.
