Abstract

(a)To characterise active site mutants of galactose oxidase to study catalysis, copper site and free radical reactivity through spectroscopic studies, including time-resolved methods, and X-ray crystallography. (b)To determine the X-ray structures of new wild-type and catalytic- intermediate forms of galactose oxidase. (c)To study the mechanism of formation of thebuilt-in cofactor involving the novel Tyr-Cys thioether bond.
