Abstract

This is a pilot study to sequence 10 cosmids from the cosmid library of the Streptomyces coelicolor A3(2) chromosome which will provide detailed costings for subsequent sequencing projects and highlight any potential problem areas. Particular issues to be addressed are: i) whether the high G:C composition of the genome (greater than 70 percent) will cause any particular problems that necessitate the use of a particular sequencing strategy; ii) whether information derived from the physical map about overlaps proves accurate at the sequencing level; iii) whether the size of the cloning vector (Supercos-1) affects the efficiency of sequencing; and iv) to determine a feasible timescale for completion of the whole sequencing project.
