Abstract
The objectives of the proposed research programme are: 1) Synthesis of oligonucleotide substrate analogues and determine their effectiveness as inhibitors of the hairpin reaction. 2) To gain further information about the three dimensional structure of the hairpin ribozyme. 3) To establish a kinetic framework for the hairpin ribozyme. The targets to be met include: 1) Identification of the best substrate analogue of the hairpin ribozyme. 2) Structural studies of the hairpin ribozyme. 3) Location of the metal ion binding sites of the hairpin ribozyme. 4) Determination of a kinetic pathway for the hairpin ribozyme.
