Abstract

(1) Target regions of the B. glabrata genome involved in schistosome resistance for local linkage and interval mapping using bulk segregant analysis (BSA) of an already extant F2 population of B. glabrata. Markers will include Sequence Characterised Amplified Regions (SCARs) and Expressed Sequence Tags (ESTs), the latter recovered from haemocyte mRNA by differential display techniques. (2) Expand the number of markers by using F2 populations from other resistant and susceptible lines of B. glabrata to broaden the survey of the genotypic basis of resistance. (3) Isolate and characterise markers and expressed sequences closely linked to, or involved in, schistosome resistance. (4) Place the evolution of schistosome compatibility in a phylogenetic framework by analysis of orthologous genes involved in schistosome resistance, identified in (1), with mitochondrial genes and the Internal Transcribed Spacer (ITS) region of rRNA genes.
