Abstract

The focus of this proposal is the development of the technology for spatially mapping adhesion forces between receptors immobilised on a probe microscope tip and their ligands on immunoassay substrates. Data will be obtained at molecular resolution in aqueous environments. Localisation of such interactions will be combined with structural imaging. The correlation of this data to parameters describing molecular binding will be investigated. Results will be related to structural transformations during unbinding. The programme exploits the analytical expertise of the University of Nottingham and Johnson and Johnson Clinical Diagnostics and the instrumental know-how of the major international probe microscope manufacturer, Digital Instruments/LOT-Oriel.
