Abstract

Rapid, automated analysis is required to characterise the heterogeneous glycosylation of many biological samples. We demonstrate high resolution HPLC and capillary electrophoresis (CE) systems in which the elution positions of fluorescently labelled glycans are predictive, so that data generated from glycan pools can be interpreted directly from one analytical run. Structures were confirmed rapidly (2 days) by a novel strategy in which aliquots of the total glycan pool were simultaneously digested with arrays of immobilised exoglycosidases and analysed by HPLC. We aim to optimise conditions for analysing femtomoles of N-, O- and charged glycans and to provide standards for extending the data bases. A computer based data handling programme will be developed to generate conclusive analyses by combining data from native glycan pools and exoglycosidase digestions.
