Abstract

Sulphoxide reductases will be studied as biocatalysts for the kinetic resolution of racemic sulphoxides. The origin of the stereoselectivity and enantiocomplementarity, shown during preliminary deoxygenation studies will be examined in depth using both whole cell and purified sulphoxide reductases. A comparison of the sulphoxide reductase- and dioxygenase- catalysed methods will provide the optimal routes to enantiopure sulphoxides of interest in the pharmaceutical sector. A study of the potential of chiral sulphoxide bioproducts as chiral auxiliaries in the chemoenzymatic synthesis of less accessible types of sulphoxides, as dienophilic reagents for determination of enantiomeric excess, and as potential oxygen-atom transfer reagents, is planned. The ability of the reductase enzyme systems to catalyse other types of stereoselective deoxygenations will be assessed. (Joint with grant 81/ABC09451).
