Abstract

Tryptase is a tetrameric proteinase implicated in asthma and inflammatory disorders. Recent structural data suggest that it should be possible to construct selective inhibitors by bridging between two or more of the adjacent active centres. This proposal is to produce and test a range of two-headed inhibitors, designed in light of the three dimensional structure, that act to bridge these sites.
