Abstract
The CD45 tyrosime phosphatase controls the signalling threshold of the T cell antigen receptor thereby playing a critical role in regulating T cell development, activation and apoptosis. In CD45-/- mice there are significant defects in thymic development and few mature T cells. The aim of this project is to introduce mutations into CD45 cDNA to lower its tyrosine phosphatase activity, screen these mutants using retroviral transfer into CD45-/- Foetal Thymus Organ Cultures and then generate a transgenic mouse line expressing a CD45 phosphatase-low mutant. The proof of concept will be a novel mouse line with a mature but non-responsive T cell repertoire, of great value in the investigation of CD45 function, TH1/TH2 differentiation and autoimmunity.
