Abstract

Glycosylphosphatidylinositol (GPI)-linked glycoproteins are an important class of membrane-anchored proteins. We are developing a strategy to select for recombinant antibodies (from an immunised phage display library) which bind exclusively to the GPI-linked and not to the soluble form of the protein. These antibodies will be extremely useful in targeting cell surface proteins such as tumour-associated antigens (PLAP and CEA), where shed antigen is a problem, in the analysis of GPI-linked proteins such as cell-surface engineering and in the general study of the topology of membrane proteins. This approach is based upon whole cell positive and reverse sections.
