Abstract
The aim of the proposed research is to examine whether cell-specific expression of heterologous Na+ transporters will increase salinity tolerance in Arabidopsis. Genes from yeast for a plasma membrane Na+ ATPase and a tonoplast Na+/H+ antiporter will be inserted into Arabidopsis downstream of a GAL4-UAS. These plants will be crossed with lines in which GAL4 (and GFP) is expressed in specific cell types, to generate a series of lines with cell-specific expression of Na+ transporters. The intracellular location of the protein product will be monitored using YFP fusions to the Na+ transporters. This range of lines will be screened for Na+ tolerance relative to the control lines, preparatory for further physiological investigations.
