Abstract

This proposal aims to address the widely appreciated need for more structural information concerning the behaviour of dynamic biological systems. To this end it is proposed to construct a dispersive fast kinetic IR spectrometer interfaced with a micro stopped-flow system. The sample volume per shot will be ca. 5-10 microlitres l compared with the 50 microlitres l currently required in the stopped-flow FTIR system we have recently developed and the deadtime will be ca. 1 ms. The spectrometer will be used to provide structural information concerning enzyme reaction and protein folding intermediates in reactive conditions.
