Abstract

This project will develop a novel approach to solving the phase problem of X-ray crystallography. Leaning on our experience in studies of selenoprotein biosynthesis, we will construct and optimise novel systems that direct selenocysteine incorporation into proteins encoded by genes cloned into a new type of vector construction. Incorporation systems for animal and insect cell lines, and for yeast, will be established and optimised. Via collaborative work we will perform proof of concept studies on two test proteins that will be subjected to crystallisation trials and X-ray analysis.
