Abstract

Two approaches will be taken to determine the macrophage-specificity of macrophage-colony stimulating factor (M-CSF). In the first approach, recombinant constructs of the M-CSF receptor (c-fms) will be prepared in which domains of c-fms are swapped with the corresponding domains from closely related receptors which are unable to support macrophage differentiation. The ability of the chimeric receptors to support macrophage growth will be established by transfection in embryonic stem cells capable of haemopoietic differentiation. In the second approach, the yeast two-hybrid system will be used to identify proteins which interact only with c-fms and not with closely related receptors.
