Abstract
Transgene expression cassettes, protected from positional effects, and under the co-operative control of all cis-regulatory elements that confer tissue specificity at a particular gene locus will be created using homologous recombination in yeast (pClasper-rescue) and E. coli (ET cloning). The well characterised Arabidopsis thaliana STM gene locus, rescued from pBAC24ol, will be the test locus. Promotorless gfp and GA 2-oxidase genes will be fused to the ATG start site substituting the STM gene, and their expression profile assayed in transgenic plants. The utility of meristem specific transgene targeting, as a tool for crop improvement, will be tested in sugar beet transformed with the GA 2-oxidase construct.
