Abstract
Stargazer mutant mice are a model of human absence epilepsy and cerebellar ataxia. A mutation in a novel voltage-calcium channel (VGCC) subunit, CACNgamma2, confers aberrant neuronal calcium signalling resulting in abnormal neurotransmitter receptor expression. Our hypothesis is that CACNgamma2 plays a role as a chaperone/adapter protein involved in trafficking, targeting, clustering and/or anchoring of the VGCC(s) it comprises to the synapse. Amino-acid sequence analysis of CACNgamma2 reveals a consensus sequence common to proteins that interact with PDZ-domain-containing proteins of the MAGUK family found at the post-synaptic density. Using the yeast-two hybrid approach we aim to identify these CACNgamma2 interactive-proteins.
